K562 cell (chronic myelogenous leukemia) line was purchased from the ATCC (CCL-243 ), cultured in RPMI 1640 (Gibco-BRL, Middlesex, UK) supplemented with 10% fetal bovine serum (PAA, Pasching, Austria). ZFN (pvax N2A-3FN FokEL-S4 WAS ZFN-L; pvax N2A-3FN FokKK-S4 WAS ZFN-R) and donor plasmids (12-18 µg) were nucleo-transferred into 1x10 6 K562 cells using the Amaxa nucleofector II and solution V (Lonza, Basel, Switzerland) applying the program T16. The bulk population was first placed under antibiotic selection for two weeks (G418 at 50 µg /ml) followed by limited dilution to isolate the two clones, K562WASKO-1 and K562WASKO-2. The cells are resuspended by pipeting up and down serveral times and transferred 1:10 to 1:5 into a new flask or plate. 6 Descripción de las características de diferenciación in vitro Description of the differentiation characteristics in vitro In the presence of PMA (Phorbol 12-myristate 13-acetate) and after 2 to 4 days some of the cells acquire the capacity to adhere to the flask and other cells become bigger, lose refraction and acquire a transparent cytoplasm.
