2 SCOPE IC BOVINE kit is a qualitative test to detect the presence of cow s milk in goat or sheep's milk. The test is an immunoassay based on the detection of bovine immunoglobulins (IgG) in milk and cheese samples. KIT COMPONENTS - 4 x Tube with 25 strip tests - 4 x Small bottle with dilution solution Please, contact Z.E.U.-INMUNOTEC or your local distributor if any anomaly is observed. SAMPLES Milk: Whole, skimmed, fresh and pasteurised milk, and whey samples can be used with this kit. Heat treatments over 72ºC/15-20 sec. can denaturise the milk immunoglobulins and give wrong results with this test. Therefore, this test should not be used with samples treated at higher temperatures. If possible, run the test before the heat treatment. Samples can be stored under refrigeration for 1 or 2 days before the assay. For longer period of time, samples should be frozen at -20ºC and thawed out to reach room temperature before running the assay. Cheese: This test should be used with cheese produced with milk treated at temperatures below 72ºC /15-20sec. Sample preparation: 1 Weigh 5 gr of an homogeneous cheese sample and add 10 millilitres of distilled water. 2 Mix with a pestle and mortar to obtain a fine homogenised sample and centrifuge at 3000 x g for 10 minutes. Alternatively, the sample can be filtered using a filter paper. 3- The supernatant or the filtered solution will be used for testing. 5

3 TEST PROCEDURE 1. Place a test tube into the rack provided. 2. Dilute the sample: - Milk samples: mix 5 drops (150 µl) of dilution solution with 1 drop (25 µl) of the milk sample and mix gently. - Cheese samples: mix 3 drops (75 µl) of dilution solution with 1 drop of the cheese extract and mix gently. Use a new disposable pipette for each sample. 3. Place the strip test into a test tube following the arrows direction. Only the white zone below the arrows must be in contact with the sample. 4. Wait for 5-10 min. 5. Read the results. NEGATIVE: A BLUE line is observed in the white central zone of the strip test. This line indicates that the test has been run properly. POSITIVE: Two lines, BLE and RED, are observed in the white central zone of the strip test. - To achieve the maximum sensitivity of the test dip strip in the simple for up to 10 min. - If a red line appears within 15 minutes, this should not be taken into account. - No lines in the white central zone means that the assay has not been run properly or that the reagents were damaged. The test must be repeated. - When hadling the strip test, take it only from the area opposite to the arrows. ASSAY PROCEDURE NEGATIVE Sample Dil. Solution Wait for 5-10 min POSITIVE 6

4 DETECTION LIMITS AND STABILITY The kit has a shelf life of 12 months and must be stored bewteen 16 and 25ºC and under dry contidions. The strips are very sensitive to humidity and must be kept inside the container provided with a desiccant bag. - Limit of Detection in milk: 0.5 1% of mixture. - Limit of Detection in cheese: 1-2% of mixture. SAFETY This kit should be used following good laboratory practices. Protective clothing is recommended while running the test. A Material Safety Data Sheet is available on request. **** IC KIT is an in vitro diagnostic test.
