(marcar@correo.unam.mx) RESUMEN ABSTRACT El objetivo de este estudio fue evaluar el efecto de la combinación de la progesterona (P 4 ) secretada por el cuerpo lúteo y una fuente exógena de P 4 (CIDR-G) en las principales variables reproductivas. Se asignaron de manera aleatoria tres tratamientos a 24 ovejas Dorset: 1) testigo, aplicación de un CIDR-G (n=9); 2) sin presencia de cuerpo lúteo (SPCL) con un CIDR-G (n=9); 3) con presencia de un cuerpo lúteo (PCL) con un CIDR-G (n=6). El CIDR-G permaneció 12 d en las ovejas. Los resultados para testigo, SPCL y PCL fueron: inicio de estro a las 43.1±4, 36±3 y 31.96±0.67 h (media±e.e; p>0.05); inicio del pico preovulatorio a las 44±6.7, 33.7±3.06 y 32±1.46 h (p>0.05); tasa de gestación 77.8, 77.8 y 83.3% (p>0.05). Se concluye que para las condiciones del presente experimento, el alterar las concentraciones de P 4 por medio de la presencia o ausencia de un cuerpo lúteo y la P 4 exógena, no afecta las principales variables reproductivas. The objective of this study was to determine the effect of progesterone (P 4 ) secreted by the CL and an exogenous source of P 4 (CIDR-G) on the main reproductive variables. Three treatments were randomly assigned to twenty-four mature Dorset ewes: 1) control group, which received a CIDR-G (n=9); 2) without the presence of corpus luteum (SPCL) plus a CIDR-G (n=9); 3) with the presence of corpus luteum (PCL) plus a CIDR-G (n=6). In all ewes the CIDR remained 12 d. The results for the control, SPCL, and PCL were: onset of estrus at 43.1±4, 36±3, and 31.96±0.67 h (mean±e.e; p>0.05); the onset of preovulatory peak at 44±6.7, 33.7±3.06, and 32±1.46 h (p>0.05); gestation rate 77.8, 77.8, and 83.3% (p>0.05). Under the condition of the present experiment, we conclude that changing P 4 concentrations by altering the presence or absence of a corpus luteum plus an exogenous source of P 4 had no effect on any of the main reproductive variables. INTRODUCCIÓN La sincronización del ciclo estral permite que un grupo de ovejas manifieste estro en determinado momento, para realizar la monta natural o inseminación artificial, agrupar nacimientos, programar destetes y vender animales por partidas. Una forma de sincronizar el ciclo estral en la oveja es mediante un tratamiento con progesterona natural usando un dispositivo intravaginal controlador de la liberación de drogas (CIDR- G) (Wheaton et al., 1993). Para usar este dispositivo en la oveja en la estación reproductiva, se coloca un CIDR- G durante 12 a 14 d, obteniéndose poco más de 90% de presentación de estros en 72 h en ovejas tratadas (Van Cleeff et al., 1998). One way of synchronizing the estrous cycle in ewes is by a treatment with natural progesterone using an intravaginal device controlling drug release (CIDR-G) (Wheaton et al., 1993). To use this device in ewes in the reproductive season, a CIDR-G is placed during d obtaining little more than 90% of estrus presentation within 72 h in treated sheep (Van Cleeff et al., 1998). The function of progesterone is to inhibit the events in the neuroendocrine axis, altering the reproductive system. A concentration higher than 1 ng ml 1 of progesterone in blood circulation hinders the pulsatile secretion of GnRH and LH (Skinner et al., 2000). 11

Una deficiencia de progesterona durante la fase lútea propicia un incremento en la frecuencia de pulsos de GnRH y LH permitiendo un prolongado crecimiento del folículo dominante, formando folículos persistentes, evitando la ovulación y comprometiendo el estado óptimo del folículo para su fertilización (Viñoles et al., 1999). Esta deficiencia en la concentración de progesterona causa una elevación prematura del pulso preovulatorio de LH, no permitiendo suficiente tiempo para el desarrollo adecuado del folículo y una deficiente formación del cuerpo lúteo al darse la ovulación (Skinner et al., 2000). La concentración y la duración de la secreción de la progesterona pueden tener una función importante en la reorganización estructural y bioquímica del folículo antes y después de la ovulación a través de una alteración en el sistema intra folicular, aunque el mecanismo no es claro (Khalid et al., 1997; Skinner et al., 2000). En ovejas la información acerca de los efectos directos del CIDR-G en los eventos endocrinos de la fase folicular después de su remoción es escasa, en la secreción, intervalo o duración de la elevación de LH (Van Cleeff et al., 1998). Por tanto, el objetivo del presente experimento fue evaluar la función que puede desempeñar la combinación de progesterona (P 4 ) secretada por el cuerpo lúteo y progesterona exógena por medio de un dispositivo liberador de P 4 en un ciclo sincronizado, así como su efecto en el inicio, duración y amplitud del pulso preovulatorio de la hormona luteinizante (LH) y fertilidad del estro sincronizado. MATERIALES Y MÉTODOS Animales experimentales El experimento se realizó del 20 de octubre al 5 de diciembre de 2001, utilizando 24 ovejas Dorset (edad promedio 3.5 años; peso promedio 56±2 kg) con una condición corporal de 3 a 3.5 en una escala de 1 a 5 (Cottle, 1991). Las ovejas estuvieron estabuladas y recibieron una dieta con 68.18% alfalfa fresca, 22.73% ensilado de maíz y 9.09% concentrado (16% PC). Sincronización de estros Los tratamientos asignados de manera aleatoria a las 24 ovejas fueron los siguientes: 1) testigo, aplicación intravaginal de 300 mg P 4 natural mediante un dispositivo liberador de la hormona (CIDR-G); 2) sin presencia de cuerpo lúteo (SPCL), utilizando un CIDR-G, con una presincronización mediante dos inyecciones de PGF 2a (15 mg Lutalyse, Pharmacia Upjohn), la primera 10 d antes de la inserción del CIDR-G, y la segunda 2 d antes de colocar CIDR-G (para evitar la presencia de un cuerpo lúteo), y 7 d después de colocar el CIDR-G se aplicó 15 mg Progesterone deficiency during the luteal phase favors the increment in the frequency of GnRH and LH surges, which allows a prolonged growth of the dominant follicle, forming persistent follicles, avoiding ovulation, and affecting the optimal state of the follicle for its fertilization (Viñoles et al., 1999). This deficiency of progesterone concentration causes a premature increase of the LH preovulatory surge, not allowing time enough for the adequate development of the follicle, and a deficient formation of the corpus luteum at the time of ovulation (Skinner et al., 2000). Concentration and duration of the progesterone secretion may have an important function in the structural and biochemical reorganization of the follicle before and after ovulation through an alteration in the intrafollicular system, though the mechanism is not clear (Khalid et al., 1997; Skinner et al., 2000). As for sheep, there is little information about the direct effects of CIDR-G on the endocrine events of the follicular phase after its removal, on secretion, interval, or duration of LH surge (Van Cleeff et al., 1998). Therefore, the objective of the present experiment was to evaluate the function that can be fulfilled by combination of the progesterone (P 4 ) secreted by the corpus luteum and exogenous progesterone, through a device releasing P 4 in a synchronization cycle, as well as its effect on the onset, duration, and amplitude of the preovulatory surge of the luteinizing hormone (LH) and the fertility of the synchronized estrus. MATERIALS AND METHODS Experimental animals The experiment was carried out from October 20 th to December 5 th, 2001, utilizing 24 Dorset ewes (average age 3.5 years; average weight 5.6±2 kg) with a body condition of 3 to 3.5 on a scale from 1 to 5 (Cottle,1991). The ewes were kept in a born and received a diet with 68.18% green alfalfa, 22.73% corn silage, and 9.00% concentrate (16% CP). Estrus synchronization The treatments, randomly assigned to the 24 ewes, were the following: 1) control, intravaginal application of 300 mg natural P 4 through a hormone-releasing (CIDR-G) device; 2) without presence of corpus luteum (SPCL), utilizing a CIDR-G with pre-synchronization by means of two injections of PGF 2a (15 mg Lutalyse, Upjohn Pharmacy), the first, 10 d before the CIDR-G insertion, and the second one 2 d before insertion of CIDR-G (in order to avoid the presence of a corpus luteum), and 15 mg of PGF 2a were applied 7 d after insertion of the CIDR-G (to guarantee the complete absence of the corpus luteum); 3) presence of corpus luteum, utilizing a CIDR-G and a previous synchronization with two injections of PGF 2a (15 mg; the 12 VOLUMEN 39, NÚMERO 1

La gestación se confirmó 60 d después del servicio utilizando ultrasonido con un transductor lineal (trans-abdominal) de 3 Mhz apreciándose la estructura ósea, columna vertebral, extremidades y estructuras carunculares de la placenta. Gestation was confirmed 60 d after the service utilizing ultrasound with a linear (trans-abdominal) transducer of 3 Mhz; bone structure, vertebral column, extremities, and caruncular structures of the placenta being visible. Sampling of progesterone (P 4 ) and luteinizing hormone (LH) Muestreo de progesterona (P 4 ) y hormona luteinizante (LH) Después de colocar el CIDR-G se recolectaron muestras de sangre vía yugular cada 48 h, para determinar la concentración de P 4 en suero sanguíneo en un periodo de 26 d. Para LH se tomaron muestras de sangre vía yugular iniciando 24 h después de retirar CIDR-G, tomando muestras cada 4 h durante 3 d. Las muestras se centrifugaron a 2500 rpm por 15 min y el suero se almacenó a 4 C para su análisis. Análisis hormonales La concentración de P 4 se determinó por radioinmunoanálisis (RIA) de fase sólida (Srikandakumar et al., 1986) con un kit comercial (Coat-A-Count Diagnostic Products Co, U.S.A.). La sensibilidad del análisis fue 0.07 ng ml 1 con coeficientes de variación intra e inter ensayo de 2.3 y 10.02%. Los análisis de LH se realizaron por RIA de doble anticuerpo (Niswender et al., 1969); la sensibilidad fue 0.78 ng ml 1, con coeficientes de variación intra e inter ensayo de 9.5 y 13.3%. El inicio y duración del pulso preovulatorio de LH se obtuvieron mediante la técnica de Van Cleeff et al. (1998) y la amplitud del pulso preovulatorio de LH según Mattioli et al. (1986). After setting the CIDR-G, blood samples were collected via jugular every 48 h in order to determine P 4 concentration in blood serum, in a period of 26 d. For LH, blood samples from the jugular were taken, beginning 24 h after removing CIDR-G every 4 h during 3 d. The samples were centrifuged at 2500 rpm for 15 min, and the serum was stored at 4 C for analysis. Hormone analyses The P 4 concentration was determined by solid phase radioimmunoassay (RIA) (Srikandakumar et al., 1986) with a commercial kit (Coat-A-Count Diagnostic Products Co, USA).The sensitivity of the analysis was 0.07 ng ml 1 with coefficients of intra and inter-assay variation of 23 and 10.02%. LH analyses were conducted by double antibody RIA (Niswender et al., 1969); sensitivity was 0.78 ng ml 1 with coefficients of intra and inter-assay variation of 9.5 and 13.3%. Onset and duration of the LH pre-ovulatory surge were obtained using the Van Cleeff technique (Van Cleeff et al., 1998), and the amplitude of the LH preovulatory surge, according to Mattioli et al. (1985). Statistical analysis Análisis estadístico Los resultados de las hembras que presentaron estro y resultaron gestantes fueron analizados con una prueba de χ 2. RESULTADOS Y DISCUSIÓN Presentación e inicio de estros La presentación de estros (100% en 3 d) no fue diferente entre tratamientos (p=0.10), resultado similar a los reportados por Godfrey et al. (1997) y Godfrey et al. (1999), quienes aplicaron un CIDR-G con 300 mg P 4 durante 12 d en ovejas. Wheaton et al. (1993) observaron 75% de estros durante 3 d en ovejas y cabras con un CIDR- S con 366 mg P 4. Los resultados de inicio de estro (Cuadro 1) no fueron diferentes entre tratamientos (p>0.05). Van Cleeff et al. (1998) observaron un inicio de estro de 35.2±1.9 h con un CIDR-G durante 8 d, colocado 10 d después de aplicar 15 mg PGF 2a, en ovejas Suffolk en estación reproductiva. Además, Godfrey et al. (1997) reportaron un tiempo de 33.6±9.5 h al inicio del estro utilizando un CIDR-G, durante 12 d en ovejas. Los resultados en el grupo testigo coinciden con el tiempo al inicio del estro de 42.4±3.4 h reportado por Van Cleeff et al. (1998) al utilizar, en ovejas, dos CIDR- G por 8 d iniciando 10 d después de la presentación de estro de un ciclo sincronizado con un CIDR-G, y al momento del retiro se tenía 8 ng P 4 ml 1. El retraso del inicio de estro se debió a una alta concentración de P 4, aunque en las ovejas testigo fue 2.3 ng ml 1 al momento del retiro, y se demoró la caída de la concentración de P 4 debido a la presencia de un cuerpo lúteo funcional. Los resultados obtenidos para el inicio del estro contrastan con los reportados por Fukui et al. (1999), quienes observaron un tiempo de inicio del estro de 21.8±1.2 h, en ovejas Suffolk, aplicando una dosis de 500 U.I. de ecg 1 d antes del retiro del CIDR-G, cuya función es proporcionar FSH y LH necesarias para la completa maduración del folículo, además de estimular la producción RESULTS AND DISCUSSION Presentation and onset of estrus The presentation of estrus (100% in 3 d) was not different among treatments (p=0.10), a result similar to those reported by Godfrey et al. Furthermore, Godfrey et al. (1997) reported a time of 33.6±9.5 h at estrus onset, utilizing CIDR-G during 12 d in sheep. The results in the control group agree with the time of 42.4±3.4 h at estrus onset reported by Van Cleeff et al. (1998), utilizing two CIDR-G in sheep for 8 d, beginning 10 d after estrus presentation of a synchronized cycle with a CIDR-G, and at the moment of removal 8 ng P 4 ml 1 had been obtained. The delay of estrus onset was due to a high P 4 concentration, though in the control sheep it was 2.3 ng ml 1 at the moment of removal, and the drop of P 4 concentration was delayed due to the presence of a functional corpus luteum. The results obtained for the estrus onset contrast with those reported by Fukui et al. (1999), who observed a time of estrus onset of 21.8±1.2 h in Suffolk sheep, applying a dose of 500 U.I. of egg 1 d before the withdrawal of the CIDR-G, whose function is to provide FSH and LH needed for the complete follicle maturation, besides of stimulating the oestradiol production required for estrus onset (Fabre and Martin, 1991). Godfrey et al. (1999) report an average time of 26.5±2.3 h for estrus onset, which is not quite understandable, because with the same procedure an estrus onset of 33.6±9.5 h was Cuadro 1. (1999) reportan un tiempo promedio para el inicio del estro de 26.5±2.3 h, lo cual es poco comprensible porque con el mismo procedimiento se encontró en ovejas de pelo un inicio de estro de 33.6±9.5 h (Godfrey et al., 1997), sin conocerse el efecto de la época del año. Inicio, duración y amplitud del pulso preovulatorio de LH En la presente investigación, 100 % de las ovejas de los tres tratamientos presentaron un pico preovulatorio de LH (Figura 1). Los resultados para inicio, duración y amplitud del pulso preovulatorio de LH (Cuadro 1) no fueron diferentes entre tratamientos (p>0.05). Los resultados para SPCL y PCL son semejantes a lo reportado por Van Cleeff et al. (1998) quienes encontraron un inicio de la elevación de LH de 33.8±1.4 h con un CIDR-G en ovejas Suffolk. El incremento de LH fue 40.4±3.1 h con un CIDR-G durante 12 d (Godfrey et al., 1999), o 42.3±1.3 h con dos CIDR-G por 8 d en ovejas Suffolk (Van Cleeff et al., 1998), resultados semejantes a 1 LH (ng ml ) Grupo testigo Horas después del retiro del CIDR-G found in hair sheep (Godfrey et al., 1997) without knowing the effect of the season of the year. Skinner et al. (2000) suggest that the P 4 dose utilized during the synchronized estrus holds up the beginning of the LH increment. The estrus onset coincides with the moment of increment of the LH preovulatory surge, which is due to an optimal P 4 concentration. Fabre and Martin (1991) injected oestradiol to ovariectomized sheep and found that the beginning of the LH increment is very close to the moment of estrus onset in sheep previously treated with P 4, and compared to sheep having received only oestradiol. Duration and amplitude of the LH preovulatory peak were not different (p>0.05) among treatments (Table 1). (2000) sugieren que la dosis de P 4 utilizada durante el estro sincronizado retrasa el tiempo de inicio de elevación de la LH. El inicio del estro coincide con el momento de la elevación del pulso preovulatorio de LH, lo cual se debe a una concentración óptima de P 4. Fabre y Martín (1991) inyectaron estradiol en ovejas ovariectomizadas y encontraron que el inicio en la elevación de LH es muy cercano al momento del inicio del estro, en ovejas tratadas anteriormente con P 4, y comparadas con ovejas que sólo recibieron estradiol. La duración y la amplitud del pico preovulatorio de LH no fueron diferentes (p>0.05) entre tratamientos (Cuadro 1). En el presente experimento la concentración de P 4, mientras estuvieron insertados los CIDR-G, no afectó la duración y la amplitud del pico preovulatorio de LH, lo cual es similar a lo encontrado por Van Cleeff et al. (1998) y Skinner et al. (2000), quienes utilizaron P 4 en ovejas en un programa de sincronización de estros. Patrón de secreción de progesterona En el tratamiento PCL se tuvo la mayor concentración de P 4 en suero sanguíneo (p 0.05), en comparación con el testigo y SPCL (Figura 2). La máxima concentración de P 4 (5 d después de insertado el CIDR-G) fue 8.6 ng ml 1 para PCL, 6.3 ng ml 1 para testigo y 4.14 ng ml 1 para SPCL. Al retirar el CIDR-G las concentraciones de P 4 fueron 2.29, 2.44 y 2.21 ng ml 1, para PCL, testigo y SPCL; en los tres grupos se alcanzó la concentración basal (<1 ng ml 1 ) 24 h después retirar el CIDR-G. No hubo diferencias entre tratamientos (p>0.05) para concentración sérica de P 4, en la siguiente fase lútea. En presencia de un cuerpo lúteo se encontró que la máxima concentración de P 4 fue 4.5 ng ml 1 para la asociación de un CIDR-S con 366 mg P 4 (Carlson et al., 1989), 6 ng ml 1 en ovejas Suffolk con un CIDR-G por 8 d y 8.5 ng ml 1 con dos CIDR-G, encontrándose al momento del retiro una concentración promedio de 4 y 8 ng ml 1 de P 4 para uno y dos CIDR-G (Van Cleeff et al., 1998). La máxima concentración de P 4 observada en ovejas del tratamiento PCL contrasta con lo reportado por Van Cleeff et al. (1998) con un CIDR-G, pero es similar a lo reportado por estos mismos autores pero con dos CIDR-G. 7 MANIPULACIÓN DE LA PRESENCIA DEL CUERPO LÚTEO EN LA SINCRONIZACIÓN DE ESTRO EN OVEJAS DORSET Tasa de gestación Los resultados (77.8% testigo; 77.8% SPCL; 83.3% PCL) no fueron diferentes(p>0.05) entre tratamientos. Estos resultados son similares a lo reportado por Crosby et al. (1991) y Godfrey et al. (1997), quienes observaron una tasa de gestación de 82.2% y 71.7% utilizando un CIDR-S con 300 mg P 4 más 500 UI PMSG, y un CIDR-G con 300 mg P 4 durante 12 d. Pero Rhodes y Nathanielsz (1988) encontraron 57% de gestación con monta natural usando CIDR-S (366 mg P 4 por 14 d) en ovejas Western en el inicio de la estación reproductiva (agosto). Ovejas que recibieron dosis bajas de P 4 tuvieron una alta concentración de estrógenos (Johnson et al., 1996; Van Cleff et al., 1998) entraron más rápido en estro (Van Cleeff et al., 1998), y pudieron ovular folículos viejos (Johnson et al., 1996) lo cual disminuye la fertilidad. En el presente experimento la alteración de los niveles de progesterona secretada por el cuerpo lúteo en combinación con una fuente exógena de progesterona no tuvo efecto en la fertilidad. CONCLUSIONES De acuerdo con las condiciones del presente experimento, el cuerpo lúteo a diferentes edades influyó en la concentración de P 4 siendo posible sincronizar la edad del cuerpo lúteo para que contribuya una mayor concentración de P 4. Sin embargo, una alta concentración de P 4 no afectó significativamente el inicio de estro, la tasa de gestación, el inicio, duración y amplitud de LH. Los resultados muestran que, posiblemente, una alta concentración de P 4 afecte más el inicio del estro. Ewes that received low P 4 doses had a high estrogen concentration (Johnson et al., 1996; Van Cleeff et al., 1998), entered estrus faster (Van Cleeff et al., 1998), and were able to ovulate old follicles (Johnson et al., 1996), which diminishes fertility. In our experiment, the alteration of progesterone levels, secreted by the corpus luteum in combination with an exogenous source of progesterone had no effect on fertility. CONCLUSIONS According to the conditions of the present experiment, the corpus luteum at different ages influenced on P 4 concentration, so it is possible to synchronize the age of the corpus luteum in order to contribute a higher P 4 concentration. Nevertheless, a high P 4 concentration did not significantly affect estrus onset, gestation rate, or beginning, duration, and amplitude of LH. The results show that a high P 4 concentration possibly might have greater effect on estrus onset.
