1 MYCOTOXINS AgraQuant Mycotoxin ELISA Test Kits Romer Labs has developed a rapid, quantitative enzymelinked immunosorbent assay (ELISA) for the analysis of mycotoxins in grains, nuts, cereals and other commodities including animal feeds. How the test works AgraQuant Mycotoxin products are direct competitive enzyme-linked immunosorbent assays (ELISA). One universal sample extraction using a 70% methanol solution can be applied to analyze six classes of mycotoxins with eight different kits. Some kits apply a dilution of the sample extract to reduce matrix effects. After combining sample extract or calibrators and enzyme conjugated toxin in the antibody coated well the competitive reaction takes min (first incubation). Subsequent washing removes unbound reactants followed by the addition of substrate starting a color reaction that takes 5 20 min (second incubation). The intensity of the color is inversely proportionate to the concentration of mycotoxin in the sample or standard. A stop solution prevents further development of the color reaction enabling reproducible measurements of the intensity using a microwell reader. Results can be interpreted by using Romer Labs spreadsheets. Kit contents Each kit comes complete with 5 to 6 standards, 48 or 96 antibodycoated microwells, 48 or 96 colorcoded dilution microwells, conjugate, substrate and stop solution. 2 AgraQuant Mycotoxin ELISA Test Read the kit insert before running the test Add 200 µl conjugate into each color-coded dilution well. Add 100 µl standards or samples to the conjugate. Mix well. Transfer 100 µl content to antibody-coated wells. Incubate for 5 60 minutes. Discard contents from the wells and wash wells with deionized water or buffer solution (5x) Tap dry the wells on absorbent paper towel. Add 100 µl substrate into each well. Incubate for 5 20 minutes. 3 MYCOTOXIN Qualitative & Quantitative Mycotoxin Test Kits Lateral Flow Devices for Rapid Analysis of Mycotoxins in various commodities. Together with Romer Labs new portable AgraVision reader and the Incubator these tests are temperature controlled and useable for on-site testing. Qualitative, designed for quick easy-to-use testing with visible detection, are available for total Aflatoxins. Quantitative test kits are designed to be used with the AgraVision and are available for Aflatoxins, Deoxynivalenol and Fumonisins. The AgraVision is a sensitive, easy-to-use laboratory instrument, measuring the intensity of the lines on an and calculating the actual mycotoxin concentration. Each kit contains 24 test strips, 24 microwells coated with the antibody particle complex (conjugate), 24 dilution wells (when necessary) and one bottle of assay buffer. 4 Quantitative Test Kit Read the kit insert before running the test as procedures for samples, swabs and rinse water are different Preheat assay buffer and AgraStrip Incubator for 30 minutes at 35 C. Remove microwell sealer and place the appropriate number of conjugate coated microwells into the AgraStrip Incubator. Dilute the sample extract with assay buffer at a ratio of 1:10, e.g. 20 µl extract plus 180 µl assay buffer. (May differ depending on kit type). Add 50 µl of assay buffer into the antibody coated wells and incubate for 30 seconds. 5 Add 50 μl of sample extract to each microwell, mixing the content in each well by pipetting it up and down 10 times. Result Interpretation A color line always appears in the upper section of the test strip to indicate that the test strip is working properly. This line is the control line. Invalid results: Valid results: 6 Put a test strip into each well. 7 Close the Incubator lid and allow strip to develop color for 3 minutes. Immediately place the strip into the AgraVision tray and analyze it with the AgraVision reader. If there is no control line in the control zone, the test is invalid and the sample should be re-tested by using a valid test strip. 2 lines are visible. 5 MYCOTOXINS WATEX Mycotoxin Quantitative Test WATEX es una solución para la cuantificación de micotoxinas fácil de usar y amigable con el medioambiente, con extracción en base acuosa para ser empleadas en análisis a campo. WATEX WATER EXTRACTION Funcionamiento de la prueba Mycotoxin Quantitative Test WATEX: Mycotoxin Quantitative Test WATEX está optimizado para la extracción de micotoxinas a partir de una muestra representativa, utilizando para ello agua destilada y bolsas que contienen el buffer soluble. Todos los kits WATEX contienen bolsas Whirl- Pak con una membrana filtrante integrada, lo que elimina cualquier paso subsecuente para clarificar el extracto. Luego de la dilución del extracto y la mezcla con el anticuerpo antimicotoxina conjugado, la muestra se incuba con una tira LFD en el Incubator. Luego de verificar la presencia de la línea control, que indica que la prueba se ha llevado a cabo en forma correcta, la tira se medirá con el AgraVision Reader. La determinación de la intensidad de las líneas en la LFD permite al AgraVision calcular la concentración de las micotoxinas en la muestra analizada. Aplicaciones Mycotoxin Quantitative Test Kits WATEX ha sido diseñado para detectar y cuantificar micotoxinas en una variedad de materias primas. Composición del kit Cada kit contiene tiras, bolsas de extracción con buffer, pocillos recubiertos con el complejo anticuerpo-partícula (conjugado), buffer de dilución, puntas de pipetas amarillas, puntas de pipetas azules, tubos de dilución, bolsas Whirl-Pak con filtros y un par de pinzas azules para analizar hasta 24 muestras de 10 g. Si se analiza bajo directivas GIPSA, con muestras de 50 g, utilizar el kit COKAS1600W que contiene bolsas grandes Whirl-Pak con filtro y buffer de extracción (COKAS1612W). 6 WATEX Leer el inserto que viene con el kit antes de llevar a cabo los análisis. Procedimiento de Total Aflatoxin Quantitative Test WATEX Precalentar el Incubator durante 30 minutos hasta que alcance los 45 C. Pesar 10 g de muestra molida y agregar un sobre de buffer en el mismo lado del filtro de una bolsa Whirl- Pak. Agregar 30 ml de agua destilada a la bolsa con filtro Whirl-Pak. Agitar vigorosamente durante 2 minutos. Dejar decantar la muestra durante 2 minutos Agregar 1000 μl del buffer de dilución a un tubo.* Tomar 50 μl del extracto de la muestra y transferir al tubo de dilución. Insertar el pocillo en el Incubator y agregar 100 µl del extracto diluido. Mezclar el contenido con pipeteando hacia arriba y abajo 10 veces. *Puede variar dependiendo del kit y la aplicación. Verificar cada aplicación. Escurrir la tira en papel absorbente y leer de inmediato en el AgraVision reader. Interpretación de los resultados Una línea de color siempre aparece en la parte superior de la tira, lo que indica que la prueba es correcta. Esta linea es la de control. Resultados no válidos: Si no hay línea en la zona de control, la prueba no es válida y se debe analizar de nuevo con una tira que funcione correctamente. Resultados válidos: se visualizan 2 lineas. La intensidad de color de la línea de prueba es indirectamente proporcional a la concentración de micotoxina y debe medirse en el AgraVision.
