Se determinó la actividad antagonista in vitro de aislamientos de microorganismos obtenidos de la rizósfera de plantas de garbanzo contra el complejo de hongos causantes de la rabia: Fusarium oxysporum, Sclerotium rolfsii y Rhizoctonia solani. Las cepas nativas que mostraron el mayor porcentaje de inhibición del crecimiento micelial de los patógenos fueron seleccionadas e identificadas como Trichoderma lignorum (CIAD ), T. harzianum (CIAD ), Bacillus subtilis (CIAD ) y Pseudomonas fluorescens (CIAD ), las cuales fueron evaluadas en campo. Las cuatro cepas y una cepa comercial de T. harzianum (T-22) fueron combinadas con Glomus intraradices, y su efectividad para controlar la rabia del garbanzo se comparó contra un tratamiento químico (PCNB) y un testigo absoluto. Los tratamientos se aplicaron a la semilla previo a la siembra y se evaluó la severidad de la enfermedad cada 15 días y la colonización de raíces por los antagonistas a los 45 días. La colonización de T. harzianum CIAD G. intraradices fue de 3.3 x 10 3 ufc/g de raíz fresca-75% y la del tratamiento de B. subtilis + G. intraradices fue de 1.3 x 10 8 ufc/g de raíz fresca-75%; mientras que en la combinación P. fluorescens + G. intraradices fue de 1.4 x 10 7 ufc/g de raíz fresca-88%. Estos mismos tratamientos presentaron una mayor reducción de la severidad de la enfermedad en 64, 57 y 51%, respectivamente, en comparación con el testigo. El garbanzo (Cicer arietinum L.) es la segunda leguminosa de grano seco que se cultiva alrededor del mundo, sólo después del frijol (Phaseolus vulgaris L.) (Hervas et al., 1998). Abstract. The antagonistic activity in vitro of microorganisms isolated from chickpea rhizosphere, was evaluated against Fusarium oxysporum, Sclerotium rolfsii, and Rhizoctonia solani, causal agents of chickpea wilt. The native strains with the higher percentage of pathogen mycelial growth inhibition were selected and identified as Trichoderma lignorum (CIAD ), T. harzianum (CIAD ), Bacillus subtilis (CIAD ), and Pseudomonas fluorescens (CIAD ). These strains and a commercial strain of T. harzianum (T-22) were mixed with Glomus intraradices and their effectiveness to reduce chickpea wilt was compared against a chemical treatment (PCNB) and an absolute control in the field. The seed was treated with the microorganisms before sowing and evaluations of disease severity were conducted each 15 days, while root colonization by the antagonistic microorganisms was assessed 45 days after sowing. Colonization of T. harzianum CIAD G. intraradices was 3.3 x 10 3 ufc/g fresh root-75% and B. subtilis + G. intraradices was 1.3 x 10 8 ufc/g fresh root-75%; while the combination P. fluorescens + G. intraradices was 1.4 x 10 7 ufc/g fresh root-88%. Chickpea (Cicer arietinum L.) is the second leguminous species of dry grain cultivated around the world, just after bean (Phaseolus vulgaris L.) (Hervas et al., 1998). In Mexico, the state of Sinaloa is the main producer of this grain for domestic consumption and export (Gómez et al., 2003). Chickpea wilt or rabies caused by the fungal complex Fusarium oxysporum Schlechtend.:Fr., Rhizoctonia solani Kühn, and Sclerotium rolfsii Sacc., is one of the main limiting factors for production; losses may range from 10 to 50% (García-Estrada et al., 2004). Some control strategies include
2 28/ Volumen 27, Número 1, 2009 En México, el estado de Sinaloa es el mayor productor de este grano para consumo nacional y para exportación (Gómez et al., 2003). Una de las principales limitantes de su producción en este estado y otras regiones del mundo, es la enfermedad conocida como marchitez o rabia del garbanzo, la cual es ocasionada por el complejo de hongos: Fusarium oxysporum Schlechtend.:Fr., Rhizoctonia solani Kühn y Sclerotium rolfsii Sacc. Esta enfermedad causa pérdidas severas en el cultivo que oscilan del 10-50% de la producción (García-Estrada et al., 2004). Algunas de las estrategias que han sido implementadas para su control son la rotación de cultivos, el retiro de socas y el uso de semilla libre de patógenos y/o tratadas con fungicidas; sin embargo, los resultados han sido poco satisfactorios. El desarrollo de plantas con resistencia genética, ofrece la mejor estrategia práctica y económica para el manejo de la enfermedad (Hervas et al., 1998); sin embargo, la presencia de nuevas razas de patógenos limita esta última estrategia (Jiménez-Díaz et al., 1993). Otra alternativa para el control de la enfermedad es el control biológico utilizando microorganismos antagonistas como bacterias y hongos. Diversas investigaciones han demostrado que el control biológico a través del uso de hongos y bacterias benéficas es una alternativa potencial al uso de fungicidas y/o fumigantes. Por ejemplo, Kaur y Muthamilan (1992) demostraron que en experimentos en campo, el complejo de hongos causantes de la marchitez del garbanzo, incluyendo Fusarium oxysporum Schlechtend.:Fr. f. sp. ciceris (Padwick) Matuo y K. Sato fue controlado efectivamente por Trichoderma harzianum Rifai. En otro estudio en semilla de garbanzo, la aplicación de la cepa RBT13 de Pseudomonas fluorescens Migula disminuyó en 52% la marchitez por Fusarium y adicionalmente incrementó la germinación, el crecimiento y el rendimiento del cultivo (Kumar y Dube, 1992). Diversas especies de Bacillus, aisladas de la rizósfera de garbanzo han demostrado capacidad para inhibir la germinación y el crecimiento de hifas de F. oxysporum f. sp. ciceris y suprimir el desarrollo de la marchitez del garbanzo (Landa et al., 1997); así mismo, las micorrizas ocasionan un incremento en la actividad de la rizósfera en favor de otros microorganismos que compiten con los fitopatógenos del suelo (Meyer y Linderman, 1986). En la selección de antagonistas para el control de la rabia del garbanzo se ha enfatizado el uso de agentes individuales; sin embargo, parece factible que incrementando el número de agentes de biocontrol con el uso combinado de diferentes especies puede resultar en tratamientos de alta persistencia en la rizósfera, con mecanismos de control diferentes y con mayor resistencia a las condiciones ambientales. Akkopru y Demir (2005) demostraron no sólo que la combinación de rizobacterias y Glomus intraradices Schenck y Smith es efectiva para controlar la marchitez por Fusarium oxysporum f. sp. lycopersici (Sacc.) Snyder y Hansen, sino que también incrementa la colonización, el peso seco y el contenido de fósforo en la planta. Barea et al. (1998) reportaron que cepas de Pseudomonas productoras del antibiótico 2-4 crop rotation, destruction of crop residues and pathogenfree seed or treated with fungicides; however, results are not satisfactory. Genetic resistance offers the best control method (Hervas et al., 1998); however, new races of the pathogen limit this strategy (Jiménez-Díaz et al., 1993). Biological control is another alternative by the use of antagonistic microorganisms such as bacteria and fungi. Kaur and Muthamilan (1992) demonstrated in field experiments that the fungal complex which cause chickpea wilt, was effectively controlled by Trichoderma harzianum Rifai. In another study, application of strain RBT13 of Pseudomonas fluorescens Migula on seed, reduced 52% wilt caused by Fusarium and increased seed germination, development and crop yield (Kumar and Dube, 1992). Some species of Bacillus from chickpea rhizosphere have the capacity to inhibit germination and hyphal growth of F. oxysporum f. sp. ciceris (Landa et al., 1997); similarly, mycorrhizae increase the activity of microorganisms which compete with phytopathogens in the rhizosphere (Meyer and Linderman, 1986). It would seem feasible to use a combination of antagonistic species with different control mechanisms and with resistance to weather conditions. Akkopru and Demir (2005) demonstrated that a combination of rhizobacteria and Glomus intraradices Schenck and Smith not only was effective for control of wilt caused by Fusarium oxysporum f. sp. lycopersici (Sacc.) Snyder and Hansen, but also increased colonization, dry weight and phosphorus content in the plant. Barea et al. (1998) reported that strains of Pseudomonas which produce 2-4 diacetyphloroglucinol increase germination and biomass of Glomus mosseae (T.H. Nicolson and Gerd.) Gerd. and Trappe, and its colonizatin in tomato (Lycopersicon esculentum Mill.) roots. A mixture of fungal antagonists like T. harzianum and vesicular-arbuscular mycorrhizae like Glomus intraradices, is effective for biological control of root pathogens in vegetable crops (Datnoff et al., 1995). The objectives of this work were: a) to isolate, select, and identify strains of antagonistic microorganisms to the causal agents of rabies, in the chickpea rhizosphere, b) to isolate and identify the causal agents, and c) to evaluate the best antagonistic strains in combination with Glomus intraradices as biocontrol agents of chickpea rabies in the field. MATERIALS AND METHODS Isolation of antagonistic microorganisms and pathogens. Chickpea roots from healthy and plants with mild infection were collected in the main chickpea-producing area of Sinaloa. Pathogens were isolated by the method proposed by Booth (1977). Roots were washed with tap water, disinfested with 5% sodium hypochlorite for 3 min, washed with sterile distilled water, and dried aseptically in an air-flow laminar chamber. Then, small pieces of root tisue were plated in potato-dextroseagar (PDA), incubated at 25 C for seven days, and the identification was carried out using the taxonomic keys of Barnett and Hunter (1998), Booth (1977), and Sneb et al. (1991). Isolation of antagonists followed the methodology by

Gerd. y Trappe, así como su colonización en raíces de tomate (Lycopersicon esculentum Mill.). La mezcla de hongos antagonistas como T. harzianum y micorrizas arbúsculo-vesiculares (MAV) del hongo Glomus intraradices han demostrado que solos o en mezcla han sido efectivos en el control biológico de fitopatógenos de la raíz en hortalizas (Datnoff et al., 1995). Estas investigaciones sugieren que las micorrizas y otros agentes de biocontrol pueden ser compatibles y usarse en combinación, lo cual induce mayor control que los agentes utilizados en forma individual (Linderman, 1988). Los objetivos del presente trabajo fueron: a) Aislar, seleccionar e identificar de la rizósfera del garbanzo cepas de microorganismos antagonistas a los patógenos causantes de la rabia del garbanzo, b) aislar e identificar los microorganismos fitopatógenos causantes de la rabia del garbanzo, y c) evaluar las mejores cepas de antagonistas en combinación con el hongo micorrizógeno Glomus intraradices para el biocontrol de la rabia del garbanzo en campo. MATERIALES Y MÉTODOS Aislamiento de microorganismos antagonistas y patógenos. La primera etapa del experimento inició con un recorrido por la zona productora de garbanzo del estado de Sinaloa. Se colectaron raíces de garbanzo de plantas sanas, y de plantas con síntomas leves que estaban localizadas dentro de los manchones característicos de la enfermedad. Los patógenos se aislaron de las raíces de garbanzo con la metodología propuesta por Booth (1977). Las raíces se lavaron con agua corriente y se desinfestaron superficialmente con hipoclorito de sodio al 5% por 3 min, se lavaron repetidamente con agua destilada estéril y se secaron en un flujo de aire laminar estéril, posteriormente pequeños trozos de tejido radicular se sembraron en medio de cultivo papa-dextrosa-agar (PDA) y se incubaron a 25 C por siete días, y para su identificación se utilizaron las claves taxonómicas de Barnett y Hunter (1998), Booth (1977) y Sneb et al. (1991). El aislamiento de los antagonistas se realizó de acuerdo a la metodología de Alexander (1994), inicialmente a las raíces se les retiró cuidadosamente el exceso de tierra, después se cortaron en segmentos de 1 cm, se tomaron 50 g de raíces y se le agregaron a 450 ml de agua destilada estéril, posteriormente se agitaron por 10 min y finalmente, se realizaron diluciones seriadas y alícuotas de estas diluciones se sembraron en medio B de King para Pseudomonas, PDA acidificado (ph 4.0 usando HCl) para Trichoderma y PDA para Bacillus. Selección e identificación de antagonistas. Se aislaron cepas de hongos y bacterias y se realizaron cultivos duales sobre PDA para seleccionar los antagonistas con habilidad de inhibir in vitro el crecimiento micelial de Fusarium oxysporum, Rhizoctonia solani y Sclerotium rolfsii. Para la evaluación de los antagonistas mediante cultivos duales se utilizó la metodología propuesta por Bell et al. (1982), la cual consistió en colocar en dos extremos de una caja Petri con PDA al Alexander (1994): Excess soil was carefully taken away from roots, then they were cut into 1 cm segments, 450 ml of sterile distilled water were added to 50 g of roots, shaked for 10 min, and serial dilutions were prepared. Aliquots were placed in King s B medium for Pseudomonas, acidified PDA (ph 4.0 using HCl) for Trichoderma, and PDA for Bacillus. Selection and identification of antagonists. After isolation of fungal and bacterial strains, dual cultures (Bell et al., 1982) on PDA were carried out in order to select antagonists with the ability to inhibit in vitro mycelial growth of Fusarium oxysporum, Rhizoctonia solani and Sclerotium rolfsii. Antagonists were placed at opposite edges of a PDA Petri plate; Bacillus and Pseudomonas were streaked and Trichoderma was placed in discs (diameter 0.5 cm) of medium with active fungal growth, and in the center of the plate a disc of medium with fungal growth (diameter 0.5 cm) of the pathogen. Checks were the pathogens without the antagonists in the plates. Petri plates were incubated at 25 C, evaluation of mycelial growth inhibition was done when the check covered the plate completely. There were five replications in a completely randomized design. Four antagonistic strains which showed the greatest mycelial growth inhibition, were selected for species identification and field evaluation. The keys of Domsch et al. (1980) were used for identification of species of Trichoderma, kit API 20 CH- Bacillus (Biomerieux, France) for Bacillus, and morphological, physiological, and biochemical test for levan, oxidase, pectolysis of potato, arginine dihydrolase, and hypersensitivity of tobacco for Pseudomonas (Schaad, 1980). Biomass increase of Bacillus was done according to Rodríguez (2005), for Trichoderma according to Serrano- Carreón et al. (2002), and for Pseudomonas according to Landa et al. (1997). Field evaluation of selected antagonists. Selected antagonists were tested in the field against the causal agents of rabies and were compared against a commercial strain of Trichoderma harzianum (T-22 ), PCNB (Pentachloronitrobenzene at 1.0 L/100 kg of seed), and an untreated check. Each biocontrol agent was mixed with Glomus intraradices (PHC-VAM-PWI ). Seed inoculation was carried out by spraying the mixtures of microorganims before sowing, at the rates indicated in Table 1. The microorganisms were suspended in 500 ml of phosphate buffer with 1 g/l of Xanthan gum (Keltrol CG-RD, Kelco Industrial Biopolymers) as adhesive. The check treatment was sprayed with only the phosphate buffer + Xanthan gum. The experiment was conducted in a field with clay soil and with previous history of heavy incidence of rabies, located at Las Tatemitas km 91.5 highway to Angostura. Seed of cultivar Blanco Sinaloa was used for the experiment. Sowing took place on December 5, crop season , and the agronomic practices followed the recommendation by INIFAP (1998). Root colonization by the biological control agents. Root colonization was evaluated in four plants randomly chosen per experimental unit 45 days after sowing. Isolation and
4 30/ Volumen 27, Número 1, 2009 antagonista a evaluar; Bacillus y Pseudomonas se colocaron haciendo un rayado con un asa bacteriológica y Trichoderma con discos de medio de cultivo con crecimiento activo de micelio (diámetro 0.5 cm), y en el centro un disco de medio de cultivo (diámetro 0.5 cm) con el patógeno a evaluar. Como testigos se colocaron los patógenos sin los antagonistas en las cajas Petri. Las cajas se incubaron a 25 C, evaluando la inhibición del desarrollo micelial cuando el testigo llenó la caja y se determinó el porcentaje de inhibición. Se realizaron cinco réplicas en un diseño aleatorizado completo. Se seleccionaron cuatro de las cepas de antagonistas que mostraron mayores índices de inhibición micelial de los patógenos para la identificación de especie y evaluación de efectividad en campo. Para la identificación de las especies de Trichoderma se utilizaron las claves de Domsch et al. (1980), para Bacillus se utilizó el kit de identificación API 20 CH-Bacillus (Biomerieux, Francia), y para Pseudomonas pruebas morfológicas, fisiológicas, y bioquímicas de levana, prueba de oxidasa, pudrición de papa, hidróxido de arginina e hipersensibilidad de tabaco (Schaad, 1980). El incremento en biomasa de Trichoderma y Bacillus se realizó en el Instituto de Biotecnología de la UNAM. Bacillus se fermentó en medio mineral con dextrosa como fuente de carbono, se utilizó un biorreactor de 43 L de volumen nominal y las condiciones fueron las siguientes: Volumen de cultivo de 30 L, aireación de 30 L/min, 30 C, 235 rpm y 24 h de cultivo (Rodríguez, 2005). Para la producción de inóculo de Trichoderma se utilizó un medio mineral y con condiciones similares a las de Bacillus, modificando solamente el tiempo a 5 días y las rpm a 180 (Serrano-Carreón et al., 2002). La cepa de Pseudomonas se incrementó en medio líquido papa-dextrosa, en un agitador rotatorio a 120 rpm a 25 C durante 48 h (Landa et al., 1997) en el CIAD unidad Culiacán, Sinaloa, México. Evaluación en campo de los antagonistas seleccionados. Los antagonistas seleccionados se utilizaron en la evaluación en campo para el control de los patógenos causantes de la rabia del garbanzo y se compararon contra una cepa comercial de Trichoderma harzianum (T-22 ), el producto de control químico comúnmente utilizado en la región (Pentacloronitrobenceno = PCNB a dosis de 1.0 L/100 kg de semilla) y un testigo no tratado. Cada uno de los agentes de control biológico fue mezclado con Glomus intraradices (PHC-VAM-PWI ), un hongo micorrizógeno con el que se trata usualmente la semilla de garbanzo. La inoculación de la semilla se realizó previo a la siembra y consistió en asperjar las concentraciones de microorganismos indicadas en el Cuadro 1. Los microorganismos se suspendieron en 500 ml de buffer de fosfatos adicionado con 1 g/l de goma Xantana (Keltrol CG-RD, Kelco Industrial Biopolymers) como adherente. El tratamiento testigo consistió en asperjar solamente buffer de fosfatos + goma Xantana. El experimento en campo se realizó en un lote de textura arcillosa con antecedentes de daño severo de rabia del garbanzo, ubicado en el campo Las Tatemitas km 91.5 de la carretera a Angostura. Se utilizó semilla de garbanzo de la variedad Blanco Sinaloa. counting of antagonists in the rhizosphere followed the methodology described by Alexander (1994). The same roots were used to determine colonization by G. intraradices; roots were subjected to clearing, staining with trypan blue in lactophenol, and the percentage of colonization was determined by the linear intersection method (Phillips and Hayman, 1970). Determination of disease severity. Evaluations were done every 15 days, from sowing to formation and pod filling, taking into consideration four central rows and counting 100 plants per row (10 cm distance between plants). The number of affected plants was recorded based on the following subjective scale: 0 = healthy plant; 1 = chlorotic plants; 2 = wilted plants, and 3 = dead plant (Carrillo, 2004). Data were used to calculate the severity index: P = ( Si x Ni)100/(3 x Nt), where: P is the severity, Ni is the number of plants with Si symptom severity, and Nt is the total number of plant per experimental unit (Landa et al., 2004). Statistical analysis. For the field experiment, a complete block randomized design was used (seven treatments and four replications per treatment), with a total of 28 experimental units, each one consisting of 16 rows, 10 m long and 0.8 m distance between rows. Variables evaluated were root colonization by antagonists and percentage disease severity. For the laboratory and field tests, mean comparison was done by Tukey s test (0.05) using the statistical package Minitab 14. RESULTS Isolation and identification of antagonistic microorganisms and phytopathogens. Rhizoctonia solani, Fusarium oxysporum and Sclerotium rolfsii were isolated directly from roots of infected plants and were maintained in PDA for later use. From a total of 35 fungal and bacterial strains present in the rhizosphere, four identified as Trichoderma lignorum (Tode) Harz (CIAD ), Trichoderma harzianum (CIAD ), Bacillus subtilis (Ehrenberg) Cohn (CIAD ), and Pseudomonas fluorescens (CIAD ), were selected based on their capacity to inhibit mycelial growth of the phytopathogens in vitro (Table 2). T. lignorum (CIAD ) showed the highest percentage of inhibition on the three pathogenic species. Root colonization by antagonistic microorganisms. All the native strains as well as T. harzianum (T-22) were capable of colonizing roots. Colonization by G. intraradices ranged from 14.7 to 88.3%. The combination of bacteria and G. intraradices and T. harzianum (CIAD ) with G. intraradices resulted in high percentage of colonization by both antagonists (Table 3). Disease supression by antagonists in the field. During the first three evaluations of severity, only the check treatment showed the highest severity and it was statistically different (Table 4). PCNB had the lowest severity level, and all the antagonists showed a reduction in the level of severity. However, in the last evaluation, PCNB showed higher disease

Colonización de las raíces por los agentes de control biológico. En cada réplica y para cada tipo de antagonista se determinó la colonización en la raíz. Esta variable se determinó en cuatro plantas seleccionadas al azar por cada unidad experimental a los 45 días después de la siembra. Para llevar a cabo el aislamiento y conteo de los antagonistas en la rizósfera se utilizó la metodología descrita anteriormente (Alexander, 1994). Las raíces que se utilizaron para determinar la colonización de los antagonistas fueron usadas también para determinar la colonización por G. intraradices; para ello, se realizó el aclareo y tinción de raíces con azul tripano en lactofenol y posteriormente se cuantificó el porcentaje de colonización por el método lineal de intersección (Phillips y Hayman, 1970). Determinación de severidad de la enfermedad. Las evaluaciones se realizaron cada 15 días desde la siembra hasta la etapa fenológica de formación y llenado de bolsa en garbanzo, tomando en consideración los cuatro surcos centrales, para lo cual se contó con 100 plantas para cada surco (10 cm de distancia entre planta y planta). En cada evaluación se registró el número de plantas infectadas considerando la sintomatología ocasionada por la enfermedad y utilizando la siguiente escala subjetiva: 0 = planta sana; 1 = planta clorótica; 2 = planta marchita y 3 = planta muerta (Carrillo, 2004). Los datos sobre la severidad de la enfermedad fueron usados para calcular el índice de severidad de la enfermedad (P) determinado como P = ( Si x Ni)100/(3 x Nt) donde: P es la severidad, Ni es el número de plantas con Si severidad del síntoma y Nt es el número total de plantas por unidad experimental (Landa et al., 2004). Análisis estadísticos. Para el experimento de campo se utilizó un diseño de bloques completos al azar (siete tratamientos y cuatro repeticiones por tratamiento), con un total de 28 unidades experimentales. Cada unidad experimental consistió en 16 surcos de 10 m de longitud y 0.8 m de distancia entre surco y surco, dando un total de 128 m 2 para cada unidad experimental. Las variables a evaluar fueron la colonización de antagonistas en la raíz y el porcentaje de severidad de la severity than the check. DISCUSSION All 35 antagonistic strains isolated from chickpea roots showed different levels of inhibition (data not shown). Strains of Trichoderma were the most effective to inhibit growth of phytopathogens. T. harzianum is the most well known species of this genus as a biocontrol agent in the soil; however, strains of T. lignorum showed the highest percentage of inhibition. Hyperparasitism was observed in the strains of Trichoderma against all the phytopathogens, since they covered partially or totally the mycelial growth, and morphological alterations of mycelia and conidia were observed. On the other hand, the effect of Bacillus subtilis and Pseudomonas fluorescens was via antibiosis, since an inhibition halo of variable intensity was produced in the contact zone with phytopathogens. The low levels of colonization by treatments T. harzianum (T-22) + G. intraradices and T. lignorum (CIAD ) + G. intraradices, suggest an incompatibility reaction. A frequently observed characteristic of mycoparasites, is that they do not always show specificity for control of pathogens; a mycoparasite may atack beneficial mycorrhizae (Whipps, 2001). Rousseau et al. (1996) reported that Glomus intraradices was highly susceptible to the attack by T. harzianum; however, they also pointed out that results were difficult to generalize, because it will depend on the aggresiveness of the strain of Trichoderma used, as it was observed with T. harzianum (CIAD ), which showed low inhibition of mycelial growth in vitro, and when combined with G. intraradices, the latter showed extensive colonization of roots. Datnoff et al. (1995) evaluated the combination of T. harzianum (T-22) and G. intraradices (PHC-VAM-PWI ) for control of Fusarium oxysporum f. sp. radicis-lycopersici W.R.
6 32/ Volumen 27, Número 1, 2009 enfermedad. Para las pruebas de laboratorio y campo, las medias fueron comparadas con la prueba de Tukey con un 5% de probabilidad de error. Se utilizó el paquete estadístico Minitab 14. RESULTADOS Aislamiento e identificación de microorganismos antagonistas y fitopatógenos. Los microorganismos fitopatógenos Rhizoctonia solani, Fusarium oxysporum y Sclerotium rolfsii fueron aislados directamente de raíces de garbanzo enfermas. Estos hongos se preservaron en PDA para ser usados posteriormente. Se aisló un total de 35 cepas de bacterias y hongos presentes en la rizósfera de garbanzo. Se seleccionaron las mejores cepas que mostraron antagonismo in vitro a los microorganismos fitopatógenos causantes de la rabia del garbanzo. Las cepas de microorganismos antagonistas presentaron diferencias significativas en los porcentajes de inhibición micelial de los fitopatógenos (Cuadro 2). Las cuatro cepas antagonistas con mayores índices de inhibición fueron seleccionadas para el experimento en campo y se identificaron como Trichoderma lignorum (Tode) Harz (CIAD ), Trichoderma harzianum (CIAD ), Bacillus subtilis (Ehrenberg) Cohn (CIAD ) y Pseudomonas fluorescens (CIAD ). Los porcentajes de inhibición estuvieron entre un 27.3 y 76.8%, F. oxysporum fue el patógeno menos inhibido en el crecimiento micelial por cada uno de los antagonistas y fueron entre 27.3 y 66.7%. La cepa de T. lignorum (CIAD ) mostró el mayor porcentaje de inhibición micelial en todos los patógenos causantes de la rabia del garbanzo. Colonización de la rizósfera por los microorganismos antagonistas. Todas las cepas nativas presentaron la capacidad de colonizar las raíces de garbanzo, al igual que la cepa comercial de T. harzianum (T-22). La colonización de G. intraradices osciló entre 14.7 y 88.3%. La combinación de las bacterias y G. intraradices resultó en una alta colonización de parte de ambos antagonistas, al igual que con la cepa de T. harzianum (CIAD ); en contraste, la cepa de T. lignorum (CIAD ) y la cepa comercial de T. harzianum (T-22) presentaron el menor índice de colonización en la rizósfera, así como también el menor porcentaje de Jarvis and Shoemaker in Florida, and 42 days after sowing, colonization by Glomus was 50%. These results show the complexity of interactions that may occur in the rhizosphere of different plants and experimental systems. The combination of rhizobacteria with Glomus in our study, rendered large populations of both antagonists. Other studies indicate that rhizobacteria stimulate colonization by Glomus sp., since they induce spore germination and promote mycelial growth. Also, they are related to changes in quantity and quality of root exudates, induced by Glomus (Akkopru and Demir, 2005; Barea, 1998). Disease severity in the last evaluation ranged from 9.64 to 17.48% and it was influenced by the antagonistic population in the roots. All antagonists reduced disease severity in the first three evaluations, but no significant differences were detected, except with the check. In agreement with the report by Díaz-Franco and Ortegón- Morales (1999), PCNB did not show disease incidence. Differences in disease severity in the final evaluation were correlated with populations of antagonists in roots; treatments with less colonization by Trichoderma spp. and G. intraradices had the highest disease severity, and no difference was detected with PCNB or the check. In the first evaluation, the fungicide treatment did not register incidence of the disease, but it had the highest severity in the last evaluation with %. Ruiz (1989) indicates that chemical treatments delay disease development, they do not reduce the final severity. B. subtilis + G. intraradices and P. fluorescens + G. intraradices reduced disease severity significantly with respect to PCNB and the check, which could be attributed to the antagonist population present in the root. These results demonstrate that rhizobacteria and mycorrhizae can coexist without adverse effects between them (Frey-Klett et al., 1997). El tratamiento químico resultó con menor nivel de severidad de la enfermedad en las primeras tres evaluaciones, de igual manera, todos los antagonistas redujeron el nivel de severidad de la enfermedad en las evaluaciones iniciales. En la última evaluación se presentaron resultados variables, ya que el tratamiento químico que inicialmente estaba protegiendo la raíz, terminó con mayor porcentaje de severidad que el tratamiento testigo. La menor severidad de la enfermedad la registraron los tratamientos B. subtilis + G. intraradices y P. fluorescens + G. intraradices los cuales fueron estadísticamente diferentes al testigo y al químico. Los tratamientos de las cepas nativas de Trichoderma + G. intraradices y la cepa comercial de T. harzianum (T-22) + G. intraradices presentaron diferencias significativas entre ellos. El tratamiento de T. harzianum (T- 22) + G. intraradices no fue efectivo en reducir la severidad de la enfermedad, ya que fue estadísticamente igual al tratamiento químico y al tratamiento testigo en la última evaluación. DISCUSIÓN Las 35 cepas de antagonistas aislados presentaron diferente nivel de inhibición (datos no mostrados). En este trabajo todos los antagonistas fueron aislados de raíces de las plantas de garbanzo que aparentemente estaban sanas o mostraban daños leves de la enfermedad dentro de los manchones característicos de ésta. Las cepas de Trichoderma fueron las más efectivas para inhibir el crecimiento de los microorganismos fitopatógenos. T. harzianum es la especie de este género más ampliamente reconocida como agente de control biológico de patógenos del suelo; sin embargo, dentro de las cepas aisladas T. lignorum presentó el mayor porcentaje de inhibición. Se observó un efecto de hiperparasitismo de las cepas de Trichoderma contra todos los microorganismos fitopatógenos evaluados, ya que Trichoderma cubrió parcial o totalmente el crecimiento micelial de los fitopatógenos, observándose alteraciones morfológicas en el micelio y conidios. En cambio, con las bacterias Bacillus subtilis y Pseudomonas fluorescens el efecto fue vía antibiosis, ya que se observó la formación de un halo de inhibición del crecimiento con diferente intensidad en la zona de contacto con los microorganismos fitopatógenos. En los tratamientos T. harzianum (T-22) + G. intraradices y T. lignorum (CIAD ) + G. intraradices, los bajos niveles de colonización micorrizógena sugieren una reación de incompatibilidad. Una característica observada frecuentemente con microorganismos micoparásitos es que no siempre presentan especificidad para controlar patógenos; un micoparásito puede tener el potencial de atacar a los hongos benéficos formadores de micorrizas (Whipps, 2001). Rousseau et al. (1996) reportaron que Glomus intraradices es altamente susceptible al ataque de T. harzianum; sin embargo, señalan que sus resultados son difíciles de generalizar, ya que esto dependerá de la agresividad de la cepa de Trichoderma, tal como lo observado Cuadro 3. 8 34/ Volumen 27, Número 1, 2009 en la cepa nativa T. harzianum (CIAD ), la cual presentó menor inhibición micelial in vitro, y cuando se combinó con G. intraradices, éste último microorganismo colonizó ampliamente la raíz de garbanzo. Datnoff et al. (1995) evaluaron una combinación de la cepa T. harzianum (T-22) y G. intraradices (PHC-VAM-PWI ) para el control de Fusarium oxysporum f. sp. radicis-lycopersici W.R. Jarvis y Shoemaker en Florida, y a los 42 días de la siembra, la colonización por Glomus fue de 50%. Estos resultados reflejan la complejidad de las interacciones que pueden ocurrir en la rizósfera de diferentes plantas y en diferentes sistemas experimentales. En este experimento, las combinaciones de rizobacterias con Glomus resultaron en altas poblaciones de ambos antagonistas. Otros estudios han reportado que las rizobacterias estimulan la colonización de Glomus sp., ya que las rizobacterias favorecen la germinación de la espora y promueven el crecimiento del micelio; además, las poblaciones de rizobacterias están relacionadas con los cambios en los exudados de la raíz tanto en cantidad como en calidad inducidos por la colonización de Glomus (Akkopru y Demir, 2005; Barea, 1998). La severidad de la enfermedad en la última evaluación varió entre un 9.64 y 17.48% y fue influenciada por las poblaciones de los antagonistas en la raíz. Todos los antagonistas redujeron la severidad de la enfermedad en las tres evaluaciones iniciales sin mostrar diferencia significativa entre ellos, sólo con respecto al testigo. El tratamiento químico no presentó incidencia de la enfermedad en la primera evaluación, esto concuerda con lo reportado por Díaz-Franco y Ortegón-Morales (1999) al evaluar fungicidas contra la rabia del garbanzo. En la evaluación final las diferencias en severidad de la enfermedad estuvieron correlacionadas con las poblaciones de antagonistas en la raíz, ya que los tratamientos con menor colonización de Trichoderma spp. así como de G. intraradices presentaron índices de severidad más altos y no mostraron diferencia significativa con respecto al tratamiento químico y al testigo. El tratamiento químico, que en la primera evaluación no presentó incidencia de la enfermedad, registró la mayor severidad en la última evaluación con %. Ruiz (1989) menciona que los tratamientos químicos retrasan el desarrollo de la enfermedad pero no reducen el nivel final de la misma. Los tratamientos B. subtilis + G. intraradices y P. fluorescens + G. intraradices redujeron la severidad de la enfermedad de manera significativa con respecto al testigo y al químico. Esta reducción de la severidad de la enfermedad es atribuida a las poblaciones de estos antagonistas presentes en la raíz. Estos resultados demuestran que dos grupos de microorganismos habitantes de la rizósfera, como son los hongos formadores de micorrizas y las rizobacterias pueden coexistir sin exhibir un efecto adverso aparente entre ellos, fenómeno que ya se ha documentado previamente (Frey-Klett et al., 1997). Además existió una relación positiva entre la colonización de raíces por los microorganismos y la reducción en severidad de la rabia del garbanzo. En estudios posteriores se podrían investigar las posibles interacciones de G. intraradices con cepas de Trichoderma spp. que presenten diferentes niveles de agresividad hacia los microorganismos fitopatógenos.
